base pairs (bp) from the C1H4 gene segment into unrelated rearrangements of V186.2. In the sample from B220lo GC cells, only a single VH V23 rearrangement encoded the YYYGS motif in CDR3, and three rearrangements, all V186.2, were not in-frame. Two of these nonproductive (nP) rearrangements resulted from incomplete codons at the V-D-J junction, whereas the third contained a complex 215-bp insertion of DNA from the V186.2 and VH 165.1 gene segments. Two hybrid sequences were present: the 215-bp insert described earlier and an in-frame exchange of 30 bp from the C1H4 exon into a productive V186.2 rearrangement. 27. C. Miller and G. Kelsoe, J. BMP-2/-4 mediate programmed cell death in chicken limb buds.
During limb development, the mesenchymal cells in restricted areas of limb bud, anterior necrotic zone, posterior necrotic zone, opaque zone and interdigital necrotic zones, are eliminated by programmed cell death. The transcripts of bone morphogenetic protein (Bmp)-2 and -4 were first detected in the areas where cell death was observed, then showed overlapping expression with the programmed ce...
متن کاملBMP signals control limb bud interdigital programmed cell death by regulating FGF signaling.
In vertebrate limbs that lack webbing, the embryonic interdigit region is removed by programmed cell death (PCD). Established models suggest that bone morphogenetic proteins (BMPs) directly trigger such PCD, although no direct genetic evidence exists for this. Alternatively, BMPs might indirectly affect PCD by regulating fibroblast growth factors (FGFs), which act as cell survival factors. Here...
متن کاملInterdigital apoptosis and downregulation of BAG-1 expression in mouse autopods
Digit formation during vertebrate limb development is a well-known example of programmed cell death. We have used this system to analyze whether the formation of the interdigital necrotic zone in mouse autopods is linked with the expression of BAG-1, a gene with an anti-death activity. Here, we demonstrate that during development of mouse autopods, BAG-1 expression is downregulated upon the ini...
متن کاملEnhancer adoption caused by genomic insertion elicits interdigital Shh expression and syndactyly in mouse
Acquisition of new cis-regulatory elements (CREs) can cause alteration of developmental gene regulation and may introduce morphological novelty in evolution. Although structural variation in the genome generated by chromosomal rearrangement is one possible source of new CREs, only a few examples are known, except for cases of retrotransposition. In this study, we show the acquisition of novel r...
متن کاملMorpho-regulation of ectodermal organs: integument pathology and phenotypic variations in K14-Noggin engineered mice through modulation of bone morphogenic protein pathway.
Ectodermal organs are composed of keratinocytes organized in different ways during induction, morphogenesis, differentiation, and regenerative stages. We hypothesize that an imbalance of fundamental signaling pathways should affect multiple ectodermal organs in a spatio-temporal-dependent manner. We produced a K14-Noggin transgenic mouse to modulate bone morphogenic protein (BMP) activity and t...
